    A triploid hybrid, which was obtained from interspecific crosses between tetraploid Primula denticulata (2n = 4x = 44) and P. rosea (2n = 2x = 22), successfully produced 11 plants by backcrossing with pollen of tetraploid P. denticulata. Analysis of ploidy level using flow cytometry and chromosome counting in the 11 BC(1) plants revealed that all progeny had much larger DNA contents and chromosome number than both parents. In this triploid-tetraploid (3x-4x) crossing, progeny was predominantly true or near pentaploid presumably produced by the fertilization between true or near triploid female gamete produced from triploid hybrid and diploid pollen of tetraploid P. denticulata. These results suggest that unreduced (3x) or near triploid female gametes were partially produced by single step meiosis, either first-division restitution or second-division restitution process. The zygotes formed by the fertilization between true or near triploid egg produced by single step meiosis in triploid hybrid and diploid pollen produced by normal meiosis of tetraploid P. denticulata might be the only survivors in embryogenesis.

    Reciprocal crossing was carried out between tetraploid Primula denticulata, section Denticulata and three taxa of P. modesta, diploid P. modesta var. fauriae, diploid P. modesta var. samanimontana and tetraploid P. modesta var. matsumurae, section Aleuritia. Inter-sectional hybrids were successfully obtained in all the reciprocal crosses except when P. modesta var. samanimontana was crossed with the pollen of P. denticulata. The number of hybrids obtained by using P. modesta var. fauriae as maternal parent was extremely low, whereas a large number of hybrids was obtained in the crosses when tetraploid species, P. denticulata and P. modesta var. matsumurae, were used as maternal parent. In all the cross combinations using P. denticulata as maternal parent, both legitimate and illegitimate crosses yielded germinable hybrid seeds. This suggested that in tetraploid P. denticulata heterostylous incompatibility might have decreased or had been lost at least on the female side. In most crosses, the DNA content of the hybrid was intermediate between that of the parents, except for the cross of P. modesta var. fouriae x P. denticulata, where the tetraploid hybrids were mainly produced due to the fertilization of the unreduced female gametes of P. modesta var. fauriae. These results suggest that in most of the zygotes with a lower ratio of female genome than male, development was inhibited in these inter-specific hybridizations.

    Inter-section hybrids were successfully obtained from reciprocal crosses between tetraploid Primula denticulata (section Denticulata) and diploid P rosea (section Oreophlomis) by applying ovary culture 4 weeks after pollination. Differences in ploidy levels were found in these hybrids by flow cytometric analysis and by counting chromosome numbers. When P denticulata was used as maternal parent, all the hybrids were triploid, with two genomes of R denticulata and one genome of P rosea. In contrast, when P rosea was used as maternal parent, two genome-type hybrids were obtained: one was a triploid hybrid, with the same genome combinations as those obtained when P denticulata was used as maternal parent, and the other type was a tetraploid hybrid with two genomes of each species. These results suggest that unreduced (2x) gametes were formed only in the female side of P rosea.

    Protoplasts were isolated from cell suspension cultures of Primula malacoides cv. 'Lovely Tokyo' and P. obconica cv. 'Aalsmeer Giant White'. P. obconica protoplasts were embedded in 0.1% (w/v) gellan gum-solidified discs comprising MS medium supplemented with 3 mg/l of 2,4-D or picloram, 0.1 mg/l of zeatin, 0.2 M glucose and 0.2M mannitol, and surrounded by a liquid medium of the same composition except for the addition of 0.1% (w/v) activated charcoal. The protoplasts formed visible colonies, which were transferred to the regeneration medium containing 30 g/l of sucrose, 0.1 mg/l of picloram and 2 mg/l of zeatin for shoot induction. P. malacoides protoplasts formed visible colonies when cultured in disc culture using 0.1%, (w/v) gellan Sum-solidified MS medium containing 5 mg/l of 2,4-D, 1 mg/l of NAA, 0.1 mg/l of zeatin and 0.4 M glucose. Small calli were transferred to MS medium supplemented with 5 mg/l of zeatin for shoot regeneration. The shoots of both species readily rooted on plans growth regulator-free 1/2 MS medium and successfully acclimatized to greenhouse conditions. The protoplast-derived plants showed some alterations in morphological characteristics from those of the in-vitro-germinated control plants. (C) 2001 Elsevier Science Ireland Ltd.     Small cell clusters with high taxol contents were selected in the following manner. Cell suspensions derived from Taxus brevifolia and Taxus cuspidata were reacted with primary polyclonal antibodies of taxol, and then labeled by magnetic or fluorescent (FITC) secondary antibodies. Cell clusters bound to the secondary antibodies were picked up manually by a small magnet or through a fluorescence microscope. It was possible to seperate cell clusters having taxol contents of more than 1000 mu g/g DW from cell suspensions which had an average taxol content of 5 mu g/g DW pr less.

    <Flower color and pigments in Primula kisoana var. shikokiana (PKS) and the related species> Flower colors of PKS were divided broadly into the categories of white, red and pinkish-purple. As a result of analysis by HPLC, 3 types of malvidin(M1,M2,M3) and 1 type of flavonol(F1) were detected from those corolla lobes, and red flowers had M2, M3 and small quantity of F1, pinkish-purple flowers had M1 and F1. Because of b^*/a^* were decreased according as the increase of F1/M1, mechanism of flower color variation in the pinkish-purple flowers were considered that originated from co-pigmentation, also the related species such as P. sieboldii.
    <Polyploidy in PKS> 4n plant and 3n plants were obtained from F_1 plants of 2n x 2n and 3n x 2n, each. And also, 4n plants were obtained by colchicine treatment to adventitious buds on root segments, efficiently.
    <Irrigation and culture methods in PKS > Drip-irrigation and Hammock culture method were good to culture, moreover root elongation and adventitious buds formation on roots were facilitated.
    <Melastoina tetramerum Hayata> M. tetramerum has little merit compared with cultivars, but, we think that it is possible to improve horticultural value of this species by interspecific cross and to investigate environmental condition in culture, especially light condition

    1.ロベリア属および近縁属植物の再分化条件などの検討:Lobelia erinusについて,長期継代カルスでは再分化能が失われていたため,新たにカルスを誘導した結果,少なくとも誘導後1年間は再分化能が維持されていることが確認された.また,Lob.loochoensisとPratia puberulaについては再分化条件が明らかになったが,Lob.chinensisについては,カルス誘導条件も確立できなかった.
    2.培養細胞由来および葉肉由来プロトプラストの効率的な培養条件の検討:Lob.erinusについて,再分化能が確認された新規誘導培養細胞を用い,プロトプラスト培養条件の検討を行った結果,浸透圧調整剤として0.1M sucroseと0.5M mannitolを組み合わせ,1〜5mg/l Picloramと0.5mg/l BAを添加した培養液を用い,Gelrite包埋培養法を用い,1〜2×10^5個/mlの培養密度で培養することによって,効率的にコロニー形成が観察されることが明らかになった.形成されたコロニーからの再分化を試みた結果,現時点でグリーンスポットが形成されており,shootが形成される可能性が高いと考えられる.また,葉肉細胞由来プロトプラストについても同様に培養を行ったが,コロニー形成頻度は低かった.
    3.子房・胚珠培養法の検討:再度,Lob.erinusとLaurentia axillrisを用い,相互交配を行い,子房培養を行った結果,Lob.erinusを子房親とした場合は子房壁由来のshootのみが形成されたが,Lau.axillrisを子房親とした場合は胚珠由来のshootが得られた.これらのshootの葉形態は,Lau.axillrisに比べ,やや丸みを帯びていたが,雑種性の確認については現在準備中である.
